相対 = relativity, but could also mean "between two persons" 性 = the nature of a person or thing, but could also mean "sex" 理論 = theory 相対性理論 = theory of relativity, but Gotland reads as "theory of sex between two persons"
Which means the easiest translation trick here is to change it to something else entirely to make the joke work. I've left it as is, but you could probably go with something like "Sexual Dimorphism" and have Gotland think that disexual was a thing or something.

The commentary makes it clear the punchline is supposed to be Gotland's "strange familiarity" of the Japanese language. So I think leaving the pun in Japanese is the best way to go.
Personally, if I needed to make this work in English I might have tried to have Gotland say "Isn't that the theory about curves causing two bodies to attract each other?" (talking about space-time curvature, of course)

Actually, 相対 can be read in two different ways with different meanings. If we read it as "soutai", it means "relative". If we read it as "aitai", it means "between two people".
Inb4 Nonco’s Iowa actually has a masters in quantum physics.
"Sei" riron is...
Theories (riron) to do with the desire and impetus for sex (sei)...?How did you reach that conclusion!?Hawawawawakakaka!?Gotland-san, can you help me study?
Easy
Theory of Relativity
Not UnderstandingI don't even know what that is...Soutaisei-riron?Soutai... I'm pretty sure that's...Are you really sure that you want to study up on lewd things!?I think Iowa-san would be better for stuff like that...Something like people agreeing to have mutual consent, right...?
