To me it would give an impression of them being Japanese... in 'slight nod to real life' way. Sort of how like Akamatsu Ken draws his character, he rarely heads into the "colorful" hair/eye territory.

They being siblings is a fan-made concept. -.-
Them being each other's reflection or "mirror images" are fan-made concepts too. Guess what else is? Character items like Miku's leek, Kaito's ice cream, or the twins' roadroller. So is Takoluka. So is pretty much anything related to the Vocaloid fandom other than names, character designs, and in some cases age. The only thing "canonical" about Rin and Len are their names, what they look like, their age, and their shared voice actress.
That's part of the whole fun. Everything is up for interpretation. If that doesn't appeal to you, why the hell do you like Vocaloid?

Look at the alternate_hair_color tag, this is simply an image of the Kagamines with black hair. The Kagenes do not only have black hair but they also have red eyes, otherwise there'd be no difference.

Though TBH, I kind of see this as Kaito being beaten by the Kagenes as first sight.
