Might be because Shujin Douji uses poisons to attack. Her NP is her dripping her alcohol to the ground which become poison, dealing damage and causing tons of debuff.

Also, her brave attack have her spewing a mist of poison right at the enemie's face.

The assassins in FGO probably also depend on their looks, at least for the females, if you look at most of them they're all wearing little to nothing or look extremely sexy.

Despite having the brute strength of an oni, some of Shuten Douji's tales included a part where he disguised as a beautiful woman to sneak up and kill his targets.

That's probably where the aspect of assassin comes in. She's also the only assassin with good physical stats, A Strength and B Endurance even when nerfed

Her toes are in the wrong order.

There's also the fact where he/she was killed thanks to a monk poisoning his/her alcohol to weaken him/her, which is most likely why Shuten's NP ingame is poisonous sake.

Otherwise, no other class really fits Shuten's mythology. I mean sure, could be a Berserker, but the way Shuten was killed and what Shuten did in stories does fit her portrayal ingame as an Assassin.
