Why is this in "No Panties?"? The only character I can see whose panty status is in any way evident is Flandre, and she's clearly naked under the open robe. Is it because Mystia's outfit is tight enough on the butt that you'd expect panty lines?

Yep. That's the whole point of the pool:

This is for images where it's very likely that the character isn't wearing panties, but you can't tell for 100% sure. Of course, this only works when she is wearing an outfit you'd normally expect someone to wear panties with, such as a schoolgirl uniform.

In this case, it's highly likely, but not 100% certain, that Misty is going regimental.

I wonder if we should tag this impending doom for Aya and the approach of Yuyuko but Mysti has helpers so maybe she is prepared

I just wasn't sure if Mysti's case was ambiguous enough to count... but then, I guess if I guessed the right person who may or may not be wearing 'em, that's a clear enough sign that they're ambiguous.
