I don't think that's right. "kyu" is a sound that's associated with Flandre's ability to break things. She has the eye of something she wants broken in her hand, she squeezes, and it breaks (though I did look it up, and it DOES have some negative connotations, but it's more about pain than death)
FOUR, however, is more commonly associated with death, since in Japanese they're pronounced the same. This tends to get referenced a lot in games (eg in the Metroid-style Castlevania games, Death always has 4444 HP, and the Death ring in Order of Ecclesia boosts most of your stats by 44 points, but renders you killable with one hit)
The reason September's being referenced, as far as I can tell, is because for some people, it means having to go back to school, or that summer holidays are over. Rather like what everyone else has said in the comments before. Since Kaguya gets the whole "NEET" thing, she's shown taunting people who have reasons to leave the house.
