Gimlet has a 'wheel' piercing on her right ear, so stallion. And so on.

As for Fine Motion... she's a special case (because of widely-circulated rumors surrounding the IRL horse), but... mare.

Also note that when the umas are wearing their gym uniforms, and if they aren't wearing pants... stallions wear gym shorts while mares wear buruma.

Yeah, the creators of the franchise actually put in a lot of research and details into making the character designs and stories, it's pretty cool.

But yeah in general, you can tell the gender of the IRL horse the Umamusume is based off of by looking at what ear their hair ornaments are, and if their gym uniforms have pants or buruma.
