Unless somehow this Pika-girl is Satoshi's mother or some sort of progenitor, I think kakkii meant for Satoshi to say「姐さん」and not 「祖さん」.

Satoshi's Pikachu is a female. So that's most likely Pikachu in human form or whatever. Unless the woman inside is actually another character from Pokemon.

ピカチューの擬人化って描いた事なかったのでちょっくら描いてみました(笑　ポケモンの年齢って良くわからないのでお姉さんにしてみました…動物は歳取るの早いって事ですしもしかしたらこんな感じではなかろうかと…そろそろマスターにならないとなサトシさん、ピカ姐さん逃げちゃいそうですよ

I hadn't drawn the personification of Pikachu before so I gave it a little go. (laugh I don't really know about Pokemon ages so I tried making her an oneesan... Animals age fast so maybe they get this kind of feeling... If you don't become a master soon, Satoshi-san, it seems like Pika-neesan is going to run away!

It's not, it's supposed to be male.

Even though Pokemon in general are meant to be "gender neutral". Pikachu is established as male.

Is was in the older episodes when Ash made a new friend who was very similar to himself. He had a male pikachu. I think his name was Richie. It was during that episode that I figured Ash's Pikachu was a female and the other guys Pikachu was a male. I was like 10 at the time though so I had probably thought that since Richie's Pikachu was a male, mainly because of it's hair and the way it acted, I figured Ash's Pikachu was female especially since the Pikachu's got along together like a couple.

Ash's Pikachu is male, because in one episode of the Pokémon anime, a female Pokémon used Attraction on some Pokémon and Pikachu was affected.
