Well, the first card Kaga played requires 5 Mana, so some number of turns has passed. Looks like it takes some (albeit overcomplicated and a bit contrived) effort to set up the combo too, I think, so it's not like one of those 'First Turn Kill' bullshit from Yugioh.

(Admittedly, I've never played Duel Masters. I just looked up some of the rules and the first few cards played.)

Edit: So the gist of it is that Kaga has a bunch of cards that let her draw more cards, or move cards from the discard zone ("graveyard") back into her hand, or play cards for 'free' (under a certain cost) from her hand or from said discard zone. She uses this to summon two key creatures (Alephtina & Marshall Crawler) and fill her hand with extra cards. She uses Marshall Crawler to put 5 cards from her hand down as shields (they sort of work like life [points]/HP in this game), bringing her shield count to 10, which apparently triggers Alephtina's instant win condition.

(I said apparently because I'm not sure of the order of resolution here, because it hinges on the instant win condition being triggered before Marshall Crawler's effect is complete, since the player is required to move an equivalent number of shield cards back into his hand after putting the extra shield cards down [in effect swapping instead adding].)
