Nope. Poison was always trans, or a "new-half" (ニューハーフ nyūhāfu), as the subject had become trendy at the time was created. The "discomfort" was over how far she had transitioned; she was (and is) officially pre-op in Japan, but American squeamishness made her post-op.
The exact gender of her palette-swap, Roxy, is not as definite. While their concept art has the word "new-half" between Poison and Roxy, Poison has long since become officially trans-female while Roxy is generally considered cis-female.
